Remembering back to the 2007 movie, at the end, Prime refers to Megatron as his "brother." There really is no followup to this, and we could have taken it either literally or figuratively.

I can't find my copy of the Transformers movie prequel comic, but I'm pretty sure the first issue even says that they are brothers in the literal sense.

So now, with Revenge of the Fallen, we find out that Optimus Prime is a descendant of the original Primes. Would this make Megatron a Prime?

 

Since Optimus is a Prime and Megatron is not, they are probably just half brothers, or maybe one of them is adopted.

 

according to rid, since transformers are not biological, two transformers are brothers if they were born at the exact same time

 

mmm I could see transformers being literal brothers if both were created from the same data base at the same time. Look at G1 Sideswipe and Sunstreaker...

I chime in every once and a while, and do a little more lurking than posting because its so tough to stay on top of it all.
