It really depends on how you distribute the weight. I think frenzy_rumble or another one of the better customizers swears by them. I was using these for my Cybertron Measor custom and the joints didn't hold up the arms in the back very well. Of course, I didn't distribute the weight as well as I thought.

It's all about placement. I have this rough Devy joint pic showing how mixmaster attached to the knee:

can you see the green plugs on E barricade? there is a refined screw which goes through the middle (hole) into barricade.

I always use screws AND apoxy resin. if the joint gets loose at all, I add a drop of super glue to the plug part.

The shoulders were similar. Same type of neo shifter joint, but with the articulated version.

My arms hold with Lego Knights ratchet joints as they don't have to support that much weight. However, if they become loose for some damn reason, you can always use superglue to give em more strength.
I use E combiner ports for the legs and they don't seem do get loose with time and hold very well.
I could've used Neo Shifter joints for the arms as they also can be fixed with superglue if they become loose.

neo-shifters have made a comeback. TRU has about 20 new toys by them. Usually the sets that are about $10 USD are a good starting point.

1 should be good, but you might want 2 if you mess up or need extras.
